چکیده

turnover, but seems unlikely as proteinase 1 is probably lysosomal (North, 19826). Nor has it been shown that substrates and inhibitors used in these studies are specific for proteinase I, which raises the possibility that other proteinases might be involved. A role for cysteine proteinases in the selective control of turnover rates of specific proteins requires more thorough investigation. It has been proposed that proteinase activity is needed during spore germination for the lysis of the spore coat. However, increases in cysteine proteinase activity occur after this process has commenced (Jackson & Cotter, 1984; North & Cotter, 1984). and it is more likely that they relate to the preparation of the cells for growth, specifically to the development of a digestive system.

Turnover of matrix proteins in mammalian mitochondria.

In cultured hepatocytes the turnover of several mitochondrial matrix proteins (e.g. acetyl-CoA acetyltransferase) appears to be initiated by CoA-mediated, sequential transformation into CoA-modified forms. This modification favours the notion that intramitochondrial degradation by a matrix-resident ATP-dependent protease may be preceded by a specific modification by CoA. In a mitochondrial matr...

Demonstration of an ATP-dependent, vanadate-sensitive endoprotease in the matrix of rat liver mitochondria.

We have recently demonstrated that mammalian mitochondria contain an ATP-dependent, vanadate-sensitive pathway for the complete degradation of mitochondrial proteins. An ATP-dependent endoprotease active against [3H-methyZ]casein and [‘4C-methyZ]globin but not ‘261-insulin has been isolated from rat liver mitochondria by a combination of ion exchange chromatography and gel filtration. This prot...

Study of the Effects of ATP Suppliers and Thiol Reductants on Toxicity of Pioglitazone in Isolated Rat Liver Mitochondria

Pioglitazone (PG) is one of thiazolidinediones used for the treatment of type II diabetes mellitus. Some reports of its hepatotoxicity exist, but the mechanism of its hepatotoxicity is not well known. In the present study, the protective effect of some ATP suppliers are investigated against mitochondrial toxicity of PG in isolated rat mitochondria. Mitochondrial viability was investigated by MT...

ATP Serves Two Distinct Roles in Protein Degradation in Reticulocytes, and One Independent of Ubiquitin One Requiring

Protein degradation in rabbit reticulocytes is a nonlysosomal process requiring ATP. Recently, appreciable evidence has been presented that ATP is required for the covalent binding of the polypeptide ubiquitin to ~-amino groups on protein substrates. To test whether linkage of ubiquitin to substrates is required for ATP-dependent proteolysis, the amino groups of 3H-methyl-casein and denatured 1...
